A hemocytometer is also known as a Haemocytometer or a cell counting chamber. It is a tool used for manual cell counting. As the name implies, the hemocytometer was originally invented for quantifying blood cells.
A hemocytometer is a device used to count cells or particles in a liquid sample under a microscope. It was originally designed for blood cell counting but is now widely used in biology and medicine to count various types of cells, such as bacteria, yeast, or other cell cultures.
Structure of a Hemocytometer
- Counting Chambers: The hemocytometer consists of a thick glass slide with two mirrored, gridded chambers. Each chamber contains a specific volume of liquid, making it easier to calculate the number of cells per unit volume.
- Grid: The counting area is etched with a grid of squares, typically 9 large squares, each of which is subdivided into smaller squares. The most commonly used grid size is a 1 mm x 1 mm square, with a depth of 0.1 mm.

- Viability Testing: By using stains like Trypan Blue, live and dead cells can be distinguished.
